Fase 1-2 dosisescalatie studie naar monotherapie IMC-F106C of combinatietherapie met checkpoint inhibitor of chemotherapie.
Intervention
IMC-F106C is een 77-kDa bispecifiek therapeutisch proteïn bestaande uit een target domein en een effector domein. Het target domein is een affinity-enhanced T-cell receptor (TCR) dat zich bindt zich aan een peptide fragment van PRAME: het HLA-A*02:01. Het effector domein is een antibody enkel string variabel fragment (scFv) wat zich kan binden aan T-cellen om zo een vrijgave van effector cytokines te stimuleren. IMC-F106C kan mogelijk een endogene antitumor response veroorzaken. • Afhankelijk van arm in monotherapy (arm A), combinatie met checkpoint remmer (atezolizumab Arm B1/pembrolizumab arm B2) of in combinatie met chemotherapie (gemcitabine arm C1, nab-paclitaxel arm C2, Caelyx arm C3).
Key outcome parameters
Bepalen van veiligheid en verdraagzaamheid van IMC-F106C.
Identificatie van maximum te toleren dosis van IMC-F106C als monotherapie of in combinatie met checkpoint remmer of chemotherapie.
Key inclusion criteria
HLA-A*02:01 positief.
PRAME positief
Progressie op standaard behandeling.

voor Arm C is eerdere behandeling met specifieke chemotherapie niet noodzakelijk.
Key exclusion criteria
Onbehandelde of symptomatische hersenmetastasen of leptomeningeale metastasen.
